Therefore, understanding the genetic diversity of different populations can be an important step towards the development and restoration of the habitat of this invaluable tree species. In this study, in order to study the genetic diversity, 10 populations including 59 genotypes via 13 RAPD primers, totally 97 alleles were generated with an average of 7.46 alleles per primer that 100% alleles were polymorphic. Polymorphic information content (PIC) varied from 0.11 (primer Oligo342) to 0.21 (primer Oligo18). Marker index criterion ranged from 0.39 (primer Oligo203) to 4.35 (primer Oligo342). Abdanan population indicated the highest value of polymorphic alleles (57.73 %) and Shannon’s index (0.251) and the highest value of unbiased expected heterozygosity index (0.172) was observed for the Banaeh population. Whereas the lowest value of polymorphic alleles (13.40 %), unbiased expected heterozygosity (0.074) and Shannon’s index (0.081) were observed for out of Zagros population. Analysis of molecular variance (AMOVA) showed that a larger proportion of genetic variation (78%) belonged to intra-populations, while 22% of genetic variation was observed for inter-populations. Cluster Analysis could not completely separate samples and showed the lack of association between molecular diversity and geographic diversity populations. The genetic diversity among the evaluated samples can be promising for restoring the habitats of P. atlantica.}, keywords = {AMOVA,He Index,P. atlantica,Genetic diversity,PCO,RAPD-PCR}, title_fa = {بررسی ساختار تنوع ژنتیکی جمعیت‌های بنه (P. atlantica subsp. mutica) با استفاده از نشانگرهای مولکولی RAPD-PCR}, abstract_fa = {گونۀ بنه (P. atlantica subsp. mutica) از باارزش‌ترین گونه‌های جنگلی ایران است که از گذشته‌های دور بهره‌برداری شدیدی از آن صورت گرفته است. شناخت تنوع ژنتیکی جمعیت‌های مختلف بنه، اهمیت زیادی برای احیا، توسعه و ترمیم رویشگاه‌های این گونه دارد. در این تحقیق، به‌منظور بررسی تنوع ژنتیکی، 10 جمعیت بنه شامل 59 ژنوتیپ با استفاده از 13 آغازگر RAPD، در مجموع 97 آلل با میانگین 46/7 آلل برای هر آغازگر تولید شد که 100 درصد آلل‌ها چندشکل بودند. محتوای اطلاعات چندشکلی از 11/0 برای آغازگر Oligo342 تا 21/0 برای آغازگر Oligo18 و شاخص نشانگر از 39/0 برای آغازگر Oligo203 تا 35/4 برای آغازگر Oligo342 متفاوت بود. جمعیت آبدانان بیشترین میزان آلل‌های چندشکل (73/57) و شاخص شانون (251/0) را نشان داد و بیشترین میزان شاخص تصحیح‌شدۀ هتروژنی (172/0) نیز در جمعیت بانه مشاهده شد؛ درحالی که کمترین میزان آلل‌های چندشکل (40/13 درصد)، شاخص تصحیح‌شدۀ هتروژنی (074/0) و شاخص شانون (081/0) در جمعیت خارج از زاگرس مشاهده شد. تجزیۀ واریانس مولکولی نشان داد که سطح بیشتری از تنوع به درون جمعیت‌ها (78 درصد) تعلق داشت، در‌حالی که 22 درصد تنوع در بین جمعیت‌ها مشاهده شد. تجزیۀ خوشه‌ای نتوانست ژنوتیپ‌ها را به‌طور کامل از هم تفکیک کند و نبود ارتباط بین تنوع مولکولی و تنوع جغرافیایی را نشان داد. با توجه به نتایج به‌دست‌آمده، با وجود در معرض خطر بودن این گونۀ ارزشمند، تنوع ژنتیکی کافی برای احیا، توسعه و ترمیم رویشگاه‌های این گونه وجود دارد.}, keywords_fa = {واژگان کلیدی: بنه,تجزیه واریانس مولکولی,تنوع ژنتیکی,شاخص هتروژنی,نشانگر RAPD-PCR}, url = {https://jfwp.ut.ac.ir/article_59039.html}, eprint = {https://jfwp.ut.ac.ir/article_59039_3d151263e01bbb8419b322fc8a8fa611.pdf} }
